Oxibendazole is synthesised through a multi-step chemical process that constructs its benzimidazole core and functional side chains. The synthesis begins with 4-hydroxyacetamide, which is alkylated using n-propyl bromide in the presence of potassium hydroxide to form a propoxy derivative. This intermediate undergoes nitration with nitric and sulphuric acids, introducing a nitro group ortho to the amide. The nitro compound is then reduced using tin(II) chloride to yield a phenylenediamine derivative. This compound reacts with S-methyl isothiourea, initiating cyclisation to form the benzimidazole ring system. Finally, the resulting 2-aminobenzimidazole is acylated with methyl chloroformate, producing the carbamate group that completes the oxibendazole molecule. | |||
