The molecule contains six stereocentres at positions C8, C9, C10, C13, C14, and C17, which define its three-dimensional configuration. Megestrol acetate is synthesised through a multi-step process starting from 17alpha-acetoxy-6-methyleneprogesterone, a modified steroidal precursor. The initial step involves dissolving this raw material in an organic solvent and reacting it with triethyl orthoformate under acidic conditions to form a protected ketal intermediate. This intermediate is then subjected to a Grignard reaction using an organomagnesium reagent, which introduces the desired side chain. Following this, the reaction mixture undergoes acidic hydrolysis, which simultaneously removes protecting groups and promotes dehydration, yielding crude megestrol acetate. The crude product is then decolorized with activated carbon, followed by recrystallisation. | |||
